Marulda solgunluğa neden olan Fusarium oxysporum f. sp. lactucae etmenine karşı Metarhizium anisoplae’nın biyokontrol potansiyelinin belirlenmesi

Abstract

Bu çalışma entomopatojen fungus olan Metarhizium anisopliae’nın marulda solgunluk hastalığına neden olan Fusarium oxysporum f.sp. lactucae etmenine karşı antagonistik etkisini belirlemek amacıyla yürütülmüştür. Marul tarlalarından alınan hastalıklı bitkilerden Fusarium oxysporum f.sp. lactucae izolasyonu yapılmıştır. Daha önceki çalışmalardan elde edilen Metarhizium anisopliae izolatlarının antagonistik etkileri in vitro koşullarda ikili karşılaştırma yöntemine göre belirlenmiştir. İn vivo koşullarda ise saksı denemeleri yapılmıştır. Yapılan denemeler sonucunda in vitro koşullarda Fusarium oxysporum f.sp. lactucae etmenine karşı en iyi yüksek etkiyi % 60.44 ile YK33 izolatı göstermiştir. Fusarium oxysporum f.sp. lactucae patojeni ile Metarhizium anisopliae’nın YK33-c izolatı arasında ise 15.12 mm’lik engelleme bölgesi oluşmuştur. Saksı denemelerinde ise en az hastalık şiddeti % 30 ile YK33-c ve Yk43-b izolatlarında görülmüştür. Kontrole göre yapılan etki değerlendirmesinde her iki izolat hastalığa karşı % 65 etki göstermiştir. Elde edilen bu sonuçlar marulda solgunluğa neden olan Fusarium oxysporum f.sp. lactucae etmenine karşı entomopatojen fungus olan Metarhizium anisopliae’nın kullanılabileceğini ortaya koymuştur.

Determination of the biocontrol potential of Metarhizium anisoplae against Fusarium oxysporum f. sp. lactucae causing wilt in lettuce

Abstract

This study was carried out to determine the antagonistic effect of the entomopathogenic fungus Metarhizium anisopliae against Fusarium oxysporum f.sp. lactucae, which causes wilt disease in lettuce. Fusarium oxysporum f.sp. lactucae was isolated from diseased plants taken from lettuce fields. The antagonistic effects of Metarhizium anisopliae isolates obtained from previous studies were determined according to dual culture methods in vitro conditions. Pot experiments were carried out under in vivo conditions. As a result of the experiments, YK33 isolate showed the highest effect with 60.44% against Fusarium oxysporum f.sp. lactucae in vitro conditions. A 15 mm inhibition zone was formed between the pathogen Fusarium oxysporum f.sp. lactucae and the YK33-c isolate of Metarhizium anisopliae. In pot experiments, the least disease severity was determined with 30% in YK33-c and Yk43-b isolates. Both isolates showed 65% effect against disease in the effect evaluation compared to the control. These results revealed that the entomopathogenic fungus Metarhizium anisopliae can be used against Fusarium oxysporum f.sp. lactucae, which causes wilt in lettuce.
