D Vitamini Reseptörü ve D Vitamini Bağlayıcı Gen Polimorfizmlerinin Atletlerde DMFT İndeksi ile İlişkisi

Abstract

Çalışmamızda kısa ve uzun mesafe koşucularda D vitamini reseptör geni (VDR) rs2228570 ve rs1544410 ile D vitamini bağlayıcı protein geni (VDBP) rs7041 ve rs4588 polimorfizmlerinin ağız ve diş sağlığı üzerine olan etkisini araştırmayı amaçladık.
Çalışmamıza 13-15 yaş arası 24 atlet katıldı. DNA izolasyonun ardından genotiplendirme gerçek zamanlı PCR metodu ile gerçekleştirildi. Ağız sağlığının tespitinde ise DMFT indeksi kullanıldı.
D vitamini reseptörlerinden FokI (rs2228570) polimorfizminin CC, CT ve TT genotiplerinin sayı ve yüzdeleri sırasıyla 9 (%37,5), 4 (%16,7), 11(%45,8), C ve T alleleri ise sırasıyla 22 (%45,83) ve 26 (%54,17) olarak bulunmuştur. BsmI (rs1544410) polimorfizminin AA, AG, GG genotipleri için sayı ve yüzdeleri sırasıyla 8 (%33,3), 13 (%54,2) ve 3(%12,5) olarak bulunurken, A ve G allelleri ise 29 (%60,42) ve 19 (%39,58) olarak bulunmuştur. D vitamini bağlayıcı protein geni rs4588 polimorfizmi için GG ve GT genotiplerinin sayı ve yüzdeleri 15 (%62,5) ve 9 (%37,5) bulunurken TT genotipine rastlanılmamıştır. G ve T alleleri 39 (%81,25) ve 9 (%18,75) olarak bulunmuştur. rs7041 polimorfizmi için CC, AC ve AA genotiplerinin sayı ve yüzdeleri 9 (%37,5), 9 (%37,5), 6 (%25) olarak bulunurken, C ve A allelerinin 27 (%56,25) ve 21 (%43,75) dir. DMFT sayıları bakımından 8 sporcu 2-4 değerine sahip iken, DMFT 10 ve üzerinde değeri iki sporcuda görülmüştür.
Kohortumuzda hem VDR ve VDBP gen polimorfizmleri genotip ve allel dağılımları açısından, hemde bu genotiplerin DMFT ile ilişkileri açısından herhangi bir benzerlik göstermemiştir. İlgili parametrelerin sporcuların ağız ve diş sağlığına olan etkilerinin daha net belirlenebilmesi için yüksek verili çalışmalara ihtiyaç bulunmaktadır.

Relationship of Vitamin D Receptor and Vitamin D Binding Gene Polymorphisms with DMFT Index in Athletes

Abstract

In the present study, we aimed to investigate the effect of vitamin-D receptor gene rs2228570 and rs1544410 and vitamin D receptor-binding protein gene rs7041 and rs4588 polymorphisms in short and long distance runners and their associations with oral and dental health.
Materials and Methods: 24 athletes aged between 13 and 15 participated in our study. Following DNA isolation, genotyping was performed by real-time PCR method. DMFT index was used to determine the oral health.
The numbers and percentages of CC, CT and TT genotypes of FokI (rs2228570) polymorphism were 9 (37,5 %), 4 (16,7%), 11 (45,8%) and 22 (45,83%) and 26 (54,17%) for C and T alleles, respectively. Numbers and percentages of BsmI (rs1544410) polymorphism for AA, AG and GG genotypes were found to be as 8 (33,3%), 13 (54,2%) and 3 (12,5%), respectively; while A and G alleles were as 29 (60,42%) and 19 (39,58%). The numbers and percentages of GG and GT genotypes for the vitamin D binding protein gene rs4588 polymorphism were 15 (62,5%) and 9 (37,5%), respectively. No TT genotype was detected. G and T alleles were found in 39 (81,25%) and 9 (18,75%) athletes. The numbers and percentages of CC, AC and AA genotypes for rs7041 polymorphism were 9 (37.5%), 9 (37.5%) and 6 (25%). C and A allelles were counted as 27 (56,25%) and 21 (43,75%). In terms of DMFT numbers, 8 athletes had low value (2-4), while only two athlete had high value (10 or above).
Our cohort did not show any similarity both in terms of genotype and allele distribution in terms of VDR and VDBP gene polymorphisms, and in terms of the relationship of these genotypes with DMFT values. There is a need for high-throughput studies to have more informative knowledge to determine the effects of relevant parameters on oral and dental health of athletes.
